Natural Bulbous Storage Trichomes Double as Phytocannabinoid Biominifactories

FACT I: In the year 2006, 41,000 metric tons of cannabis yielded approx. 100 metric tons of hashish (the balance being left to be consumed as herb) as opposed to 7,000 metric tons of opium yielding 606 metric tons of heroin and 12,000 metric tons of coca leaf yielding 984 metric tons of cocaine per the United Nations World Drug Report of 2007.

FACT II: THCA acid synthase (THCAS) is a secreted biosynthetic enzyme of the cannabis plant.

The (THCAS) sequence contains a 28 amino acid signal peptide and eight putative N-glycosylation sites.

Recently, it has been discovered that the storage cavities of the cannabis trichome(s) are not simply the primary site(s) for the accumulation of phytocannabinoid(s), but are also the primary site(s) where biosynthesis of the THCA synthase molecules do occur.

It has been shown, the resulting THCA synthase and the precursor substrate CBGA are both toxic apoptosis inducing defense terpenophenolic compounds, and as such, are biosynthesized and stored away from the stoma cells of the plant within the bulbous glandular trichome cannabinoid biominifactories.

If an insect attempts to nibble at a cannabis flower bud, it will taste the primary and secondary defense metabolites of cannabis (including hydrogen peroxide) and either die or leave very quickly.

Because of the elevated manufacture and storage of the cannabinoid metabolites within the bulbous trichomes, the stoma cells of the plant are unaffected by the twin primary by-produced defensive molecules of hydrogen pyroxide and psychoactive THCA synthase, as well as the other 66 or so cannabinoid variations produced by the plant such as cannabidiolic acid (CBDA), cannabichromenic acid (CBCA) and cannabigerolic acid (CBGA).

Synopsis:

Tetrahydrocannabinolic acid (THCA) synthase, a supposed FAD-dependent flavo-proteic oxido-reductase enzyme present in Cannabis sativa L., oxidizes cannabigerolic acid (CBGA) via a mechanism similar to that of the berberine bridge enzyme (BBE) that occurs naturally in the alkaloid biosynthesis of Eschscholtzia californica.

Both plant mechanisms need an attached flavin cofactor to proceed, in addition to an absolute requirement that molecular oxygen be present for the reaction to occur.

The THCA synthase reaction in Cannabis sativa L. can be summarized, as follows, after production of the substrate cannabigerolic acid (CBGA) has occurred in the plant from resident geranyl pyrophosphates and olivetolic acids;

While acting upon (ie. oxidatively cyclizing the mono-terpene moiety via hydride transfer) of the substrate cannabigerolic acid (CBGA) when the required molecular (O2) oxygen is present, the covalently flavinylated oxidase enzyme tetrahydrocannabinolic acid synthase (THCAS) which, by the way, has been cloned via reverse transcription, will produce the precursor molecular substance known as tetrahydrocannabinolic synthase (THCA) + H2O2 (hydrogen peroxide, as a byproduct of the reaction) when assisted by a currently unidentified attached proprietary flavin cofactor, perhaps an additional covalently ancient cross adapted inherited bacterial flavinylated enzyme.

The resultant precursor molecular substance tetrahydrocannabinolic synthase (THCA) can then be non-enzymatically decarboxylized into pure tetrahydrocannabinol (THC) and consumed either via heating in a vaporizer or by smoking, or by eating.

The bottom line:


Genetic over-expression of tetrahydrocannabinolic acid synthase (THCAS) will produce tetrahydrocannabinolic synthase (THCA) rich plants with increased therapeutic potential.

WARNING: Your liver will convert the pure tetrahydrocannabinol (THC) if ingested via plant matter into an 11-hydroxy-delta-9-THC molecule chain. This 11 molecule chain is four times more potent than the direct tetrahydrocannabinol (THC) delivered via inhalation of pure or mixed vapors. Further, there is a lag time between eating the plant matter, and feeling the effect, which is an accumulative effect if consumption is continued.

Non-Psychoactive CBD Stops AD Progression in Swiss Cheese

Cannabidiol (CBD = C21+H30+O2) is the principal non-psychoactive component of the natural phytocannabinoid plant factory known as Cannabis sativa, or hemp.

(CBD) has been found to possess potent anti-oxidant properties in the laboratory that offer neuroprotection against glutamate toxicity, and may therefore be therapeutically beneficial in curing Alzheimer’s Disease (AD).

Since (CBD) has empirically proven the ability to prevent the deleterious effects of the plaque (Ab) and also to ameliorate several key features of (AD) pathology, including tau hyperphosphorylation, oxidative stress, neuroinflammation and apoptosis, it is a therefore given not to force farmers to stop their dairy cows from feeding on such low (THC), high (CBD) concentrations of live hemp as are found in the country of Liechtenstein.

Alzheimer’s Disease is the leading cause of dementia among the elderly.

President Ronald Reagan (1980 – 1988), a staunch anti-drug Republican governor from the State of California and stifling supporter of the DEA

was the poster child for this debilitating disease.

Ironically, while in office, his “Just Say No” campaign against non-pharmaceutical embraced advancements paralyzed the progressive movement towards the discovery of new therapies for both

Alzheimer’s Disease and AIDS.

With the ever-increasing size of the over 50 population, cases of Alzheimer’s Disease are expected to triple over the next 10 to 20 years.

Consequently, the development of treatments that slow or halt the progressions of the disease have become imperative to both the improvement in the quality of life for patients and also to reduce the health care costs attributable to Alzheimer’s Disease.

The phytocannabinoid delta 9-tetrahydrocannabinol (THC) is naturally occurring in nature having been found to be the primary molecule of activity present in the cannabis trichome.

This naturally occurring molecule competitively inhibits the enzyme acetylcholinesterase (AChE) as well as prevents (AChE) induced amyloid peptide (A) aggregation, the key pathological marker of Alzheimer’s Disease.

Computational modeling of the (THC) – (AChE) interaction revealed that (THC) binds in the peripheral anionic site of (AChE), the critical region involved in the generation of amyloids, a process known as amyloidogenesis.

(THC) is, therefore, a considerably superior inhibitor of (A) aggregation as compared to currently approved drugs prescribed for the treatment of Alzheimer’s Disease.

This study provides a previously unrecognized molecular mechanism through which endo and phyto cannabinoid molecules may directly impact the progression of this debilitating disease.

Eric Steenstra, President of the North Dakota Vote Hemp Initiative (VH), a non-profit organization working to bring industrial hemp farming back to the United States (Case No. 4:07-cr-042) recently lamented the US District Court’s “moot” decision against the embattled registered hemp farmers of the State of North Dakota.

 


“We feel that the lower court’s decision not only overlooks Congress’s original legislative intent, but also fails to stand up for fundamental states’ rights against overreaching federal regulation.

 

 


[The country of] Canada grows over 30,000 acres of industrial hemp annually without any law enforcement problems. In our federalist society, it is not the burden of the State of North Dakota’s citizens to ask Congress in Washington, D.C. to clear up its contradictory and confusing regulations concerning Cannabis. [Rather], it is their right to grow industrial hemp pursuant to their own state law and the United States Constitution,” steemed Steenstra.

Phytocannabinoid (THC) Directly Induces Apoptosis In Cancer Cells by Disrupting Cancer Cell Mitochondria Sans Endocannabinoid Receptors (CB1) or (CB2)

 

 


Time-lapse microscopy of human lung cancer (H460) cells showed that the endogenous cannabinoid anandamide (AEA), the phytocannabinoid Δ-9-tetrahydrocannabinol (THC) and a synthetic cannabinoid (HU 210) all caused morphological changes characteristic of apoptosis.

 

Janus green assays of (H460) cell viability showed that (AEA) and (THC) caused significant increases in (OD 595 nm) at lower concentrations (10–50 μM) and significant decreases at 100 μM, whilst (HU 210) caused significant decreases at all concentrations.

In rat heart mitochondria, all three ligands caused significant decreases in oxygen consumption and mitochondrial membrane potential.

(THC) and (HU 210) caused significant increases in mitochondrial hydrogen peroxide production, whereas (AEA) was without significant effect.

All three ligands induced biphasic changes in either mitochondrial complex I activity and/or mitochondrial complex II–III activity.

These data demonstrate that (AEA), (THC), and (HU 210) are all able to cause changes in integrated mitochondrial function, directly, in the absence of endocannabinoid receptors.
